I've to extract RNA of specific protein from blood using Zymo Research kit to investigate the expression of that protein, so would any one please tell me which way is best for extract RNA is it from whole blood or from mono-nuclear cells ?

I would ask you about the expression of RNA in population. which cell type express your desired protein in excess. If your protein is expressed in Granulocytes than there is no way to opt for PBMCs.
But if your protein has higher expression in mono-nuclear cells than granulocytes, go for PBMCs.
if the desired protein is non specifically expressed in all blood cells, than go with whole blood, as Granulocytes has much expression profile than lymphocytes. you will get more total RNA from whole blood rather than PBMCs.
Jitendra Kumar Shandilya thx. this protein didn't study as protein expression in blood just as RNA, and it's Low lineage specificity it's detected in all.
