Senescence, defined as the arrest of growth of mammalian cells and prevents unlimited proliferation is of considerable interest in cancer treatment.
Any compound that induces DNA damage should do the trick. If they do not undergo cell death then they should enter cell senescence.
Ionising radiation, hydrogen peroxide etc all work well.
We have found that, in wild-type p53 cell lines, you can augment the effect of ionising radiation by pre-treating with compounds such as Nutlin-3. Nutlin-3 inhibits the interaction of MDM2 and p53, leading to increased p53 stabilisation, which in turn appears to lead to senescence. About 5 uM will do it for head and neck cell lines.
