To know polarity defect (by knocking out some specific genes) I am looking for a breast cancer cell line that has high polarity, forms tight junctions and easy to transfect.
Although I don't know of a breast cancer cell line that would ideally match your characteristics, I would look into MDA-MB-231 and MCF-7. Both are highly characterized (MDA-MB-231 I believe is polar Article Untargeted polar metabolomics of transformed MDA-MB-231 brea...
), and both are known to form tight junctions (

Both cell lines are likewise easy to transfect (see this reagent for MDA-MB-231: https://altogen.com/product/mda-mb-transfection-reagent-breast-adenocarcinoma/ and this one for MCF-7: https://altogen.com/product/mcf-7-transfection-reagent-breast-cancer-htb-22/). There are probably other cell lines that might better work for your research, but I wouldn't know which.
