I wish to treat the murine endothelial cell lines with some factors to check the pro-inflammatory cytokine release. I read some papers and found different treatment strategies. Is it important to incubate the cells for serum starvation (1% FBS+DMEM) prior to the treatment? If yes, then for how long (24 hr?) and how would it be different if I treat the cells in complete medium?

If there is a potential chance of getting endogenous cytokines in the serum then I will suggest to make the cells serum starved. But it is quite unlikely to get a decent amount of cytokines in the commercially available heat inactivated serum as those are usually dialysed extensively to remove hormones, cytokines, glucose, amino acids, and many other components.
