I recently started growing HUVECs to study the angiogenic and inflammatory response wrt certain treatments. I am using EBM-2 complete medium (2% FBS and growth factors including exogenous VEGF) by Lonza.
My target is to measure the expression of certain inflammatory/ angiogenic molecules and adhesion markers, at both RNA and protein level.
My main question is: In my case, what are the appropriate conditions for the treatment of HUVECs? The treatment length will be upto 48 hours.
I have basal EBM-2 (no growth factors, no serum) for this purpose. But, I am not sure how long cells would survive in the absence of serum.
Any advice is highly appreciated.

Would it make sense to use EBM-2 basal medium supplemented with low serum (e.g. 0.1% FBS, or 0.1% BSA) without any growth factors, to be used for the treatments and viability assays?
Is hydrocortisone and Ascorbic acid absolutely needed?
In our hand Huvecs work best in shorter period of time . Up to 18 hrs the best with any treatment so My suggetion is just do the experiments till 24hrs after that Huvecs all the cells start dying.
