I have seen within a number of different papers a big range of concentrations PMA utilised for THP1 differentiation. Some use quite weak conc and wait a few days for differentiation while others use quite strong conc for overnight change.
I have 1MG PMA from sigma. I was simply going to make a 1mg/ml solution in DMSO and add 1ul of the stock to each 10 ml media, giving a final working conc of 100 ng/ml.
This seems to be around the middle of the scale. Anyone having success with THP-1 cells?
Was you trying to differentiate it in macrophages? Have you checked the differentiated state of THP-1? I'm with the same doubts about the concentrations and times and way to confirm that the differentiated cells are macrophage like cells. Have you succeed?
