Most of membrane proteins require their lipid enviroment to maintain their active conformations (for example, g-coupled receptor, or many transmembrane transporters). Sometimes, the binding domain is located in the extracellular domain, without requiring the membrane domain, so you can immobilize only the ECD (recombinant). It depend also by the degree of sensitivity you want to achieve. The concentration of your receptor in the membranes could be small, so it could be difficult to study the interaction with small molecules or medium size proteins.
