We are working with colon cancer cell lines (HT29, HCT116, SW480) in our lab, and we observe some changes in procaspase-3 expression when 15-LOX-1 enzyme is overexpressed. However, the apoptosis inducers we tried (Actinomycin-D, Doxorubicin) did not show a convincing induction of apoptosis, therefore we had to perform our experiments without a positive control.
Is there an apoptosis inducer that you can suggest to be used as a positive control for these cells?

You can actually use many agents. To name a few, you can use paclitaxel or 5-FU.

We have tested Bortezomib (5-100 nM) on HCT116 cells, after 48h treatment we have more than 60% of cells with cleaved caspase 3.
