Can anyone suggest transfection methods for big inserts in neuroblstoma or glioblastoma cell lines?

Does anybody know why the transfection of alpha-fodrin (250 kda) is so less efficient in cell lines like glioblastoma U251 and neuroblastoma IMR 32? It works well in HEK293. I have tried both...
