I'm working on the role of Vitamin D on HIV infection.
Classically, the FBS-supplemented RPMI medium has been broadly used but it contain wide amounts of Vitamin D so that we are looking for a suitable serum-free medium for culture PBMCs, monocytes, dendritic cells, CD4+ and CD8+ T cells and NK cells, which are the cells target of most of our analyzes.
Our procedures include standard culture of the above mentioned primary (and sometimes monocyte and lymphocyte-derived cell lines) cells, study cell responses after polyclonal stimulation (CD3/CD28, PMA/Ionomicyn, PHA, TLR-agonists, Cytokines) and after antigen specific-stimulation.
Some recomend X-VIVO 15 (lonza), others recomend LGM-3 (lonza), others recomend OpTmizer™ CTS™ T-Cell Expansion SFM (Invitrogen), and others AIM V (Gibco). any recommendation?
I used X-vivo 15 for culturing dendritic cells in serum free condition and it worked very well.
