I am so interested in the topic of UPR; I read some works of master researchers. I have a question; approximately in all UPR studies, researchers claim UPR phenomenon impulse cell to release some materials (like cytokines, etc) or primordial changes (alternative gene expression)(I believe cancer cells utilize UPR to reach dedifferentiation), interestingly, all cells using in UPR studies was so typical and predictable like cell lines or even primary cell, as we know these cells are sensitive to harsh situation. I think using Amniotic cells can open a new way in UPR context. because they constantly face a range of situations. question is; in your view is there any difference in Amniotic cells vs other cells in case of UPR?

Treat anamniotic cell with a combination of nelfinavir and Celecoxib and you will find the same unfold protein response and ER stress yout find in a cancer cells.

each of harsh situation ingredients can execute UPR, e.g. Amniotic (stem) cell constantly put up with a range of Amniotic fluid components which can ignite UPR but they work properly. how do you interpret this? may sensitizing dose (shocking dose) is the candidate which has the potential to offer this exception! if dose concept is correct I wonder that UPR response pathways in these two sorts of cells are same (may there are more UPR pathways than illustrated before, or there is blocking shortcut or something)!

It is only a matter of how much unfolded protein is produced. ER has a threshold for coping with unfolded protein. Once that threshold is surpassed ER stress develops. UPR is only part of the ER reaction to this stress. It is a phylogenetically conserved reaction because you can find it in all species starting with yeast and ascending in the biological ladder. All the cells whether normal or malignant have the ability to respond with UPR when unfolded proteins or defective proteins accumulate in ER. The UPR is basically always the same, whether a yeast or a human cell. If UPR does not show up in a certain situation it is because there is no ER stress. In other words, the amount of accumulated protein in the ER did not reach the stress threshold.
