We have several essential oils we want to test in cell culture for cell viability using XTT assay. We only have 500 ul or less of each oil. We want to test these oils at concentrations that range around 10-200 ug/ml. Considering the maximum concentration of DMSO tolerated by cells (0.1%), how much oil and DMSO do we need to prepare our stock solution? Could somebody explain to us how to proceed step-by-step?

Dear Chiara,
Most cell types will tolerate 1% DMSO, or even 2%. Using DMSO at 1% in your final cell culture then your solution of oil needs to be 100x more concentrated, ie 20 mg/ml dosed at 1% gives a concentration of 200 ug/ml. Assuming a specific gravity of 1 (actually likely to be
