I want to do some MTT assays using Mouse 3T3 fibroblasts and do not know the suitable cytotoxic drug which should be ideal for 3T3 cells as a positive control. Any suggestions will be appreciated.

Actually at least a quercetin does not have very good solubility and able to reduce MTT by self without cells. Mentioned quercetin IC50 is equal to 6 ug/ml and under this conditions precipitation of one could occurs. Thus this limitation could account to false reduction of MTT even after washing step. So, if you are looking for positive (toxic) control just add to well 30 % of ethanol or 1 % SDS.
