Can we use just only universal primer for invertebrates or do we need to develop or design primer for certain classes or group for the amplification method to be successful?
The "universal" COI primers work for many inverts, but not for all. You should scan the literature for the right primers. There are plenty of papers on flukes and nematodes. Primers are cheap to purchase. If you already have the universal primers, you could try them on some representative specimens and see what results you'll get. That'll just cost you one PCR run + one gel or two. Regardless what primers you have, you should probably do that for all of them. You could of course develop your own primers, but that work may have already been done.
