As I am a beginner every time i faced the smearing or uneven bands on gel. What all the factors affecting that affect DNA gel electrophoresis procedure?
A picture would help and is the dna whole genomic dna or a pcr product please?
Wavy samples can often be a result of too much salt in the sample and too much dna. Try loading less and running at half the voltage that you are currently using and make sure that you have given the gel plenty of time to set and cool down before using it
Here are some factors affecting that affect the DNA gel electrophoresis procedure:
1. Agarose concentration
2. DNA loading volume
3. DNA quality and concentration
4. Voltage and run time
5. Buffer choice and pH
6. Gel size and thickness
7. Running temperature
8. Staining and visualization.
By optimizing these factors and carefully controlling experimental conditions, you can enhance the accuracy and reliability of DNA gel electrophoresis results.
with genomic dna we do not get bands on the gel but the degraded dna will form a smear down most of the sample track with a strong signal not far from the well of high molecular weight dna. If your next step is a pcr thn I would not worry about degraded dna as it will amplify ok
Degraded Genomic DNA gives a smear. If you want to have sharp band please have a look for your extraction protocol and work on the handling of samples.
