Hello, I was wondering if there is any literature out there, or personal experience on the derivation efficiency of mESC lines from aged (7-8 months) oocytes/ blastocysts..

I am seeing a stark difference in my young and old models' ES isolation efficiency data.. but because I started my work with young mice (as getting young and old mice at the same time wasn't an option then), so my efficiency could have influenced the results.

Also, is there any possibility- statistically- considering time points of the experiments performed, to negate the self efficiency factor which could have a large contribution to the results in this particular case.

I could of course relate my findings to the ICM:TE ratio in the blastocysts of each model, and see if I get any relation with my results.. But is there any literature that might support/ contradict these findings?

Many thanks.

Cheers!

Pooja.
