What is the recommended dose of insulin to perform insulin tolerance test in C57BL/6JUnib mice (fed on chow or high-fat diet)? I have found 0.1 to 10 UI/kg in the literature. Which is an adequate fasting time?
Since rodents are nocturnal animals, overnight fasting is not recommended since it causes starvation. Animals should be fasted for 6 hours in morning say 8 a.m to 2 p.m.
Our laboratory does 0.75-0.80 UI/kg body weight. We do not fast. Many of our colleagues do a 6 hour fast in the morning from 8-2 (which we do for GTT's). However, we have found that even with this short fast and relative small insulin concentration, many mice go hypoglycemic and need withdrawn from the study. Particularly the young, insulin-sensitive ones.
