Hi guys, After 72 hours of transduction of Adeno to Hec cells the color of the medium change to purple, cells have been de-attached and many tiny particles are seen. Have you had any experience about it?
That sounds like a lytic infection is occurring. Hec is a human endometrial adenocarcinoma cell line. Adenovirus is used as a vector for gene transduction not relying on either host cell replication or viral genome integration. It does replicate via a lytic infection. Some adenoviruses have been engineered to be oncolytic. Are you using a normal adenovirus strain or one that has been engineered? It is possible that the virus you are using is either replicating in Hec cells and thereby lysing them, or that the protein the virus you are using induces lysis in Hec cells.
