I used CMC congo red agar for the screening of cellulase using Aspergillus terreus. From the plate it is found that there is a clearance of CMC and with that there is a blue color development (see the figure). While using other microorganisms like Fusarium sp., Mucor sp., and Rhizopus sp. they didn't show any such blue coloration. Any idea? Thank You in advance.

2. is it after theaddition of HCL. If so it is due to the change in pH of the medium.
3. If, not hope you have the direct answer that the medium has become acidic as we understand the end product of a fungal culture is acidic always.

From above I conclude, Aspergillus terreus produce high amount of acidic metabolites than other fungi. May be Sir the cellulase enzyme produced was acidic in nature??
may be true. However, we have to confirn the same by studying its actoivity at different pH range.

I agree with both answers, indeed, Congo red is an acid-base indicator, and decreasing pH the color changes from red to violet. It is very useful when doing zymogramm by SDS-PAGE. Congo red does not provide a good contrast, and if we immerse the gel in an acidic solution is very useful to see the bands of the enzyme hydrolysis.
Usually cellulases works well in acidic conditions, in a pH of 3 to 7.5 or 8 ... is not well known an alkaline pH Cellulase activity.
