I am working on cancer cell lines and want to check the effects of TSA on HDACs. Some article has suggested to treat the cell lysate with TSA and check the HDAC activity but we want to check the effects of TSA on HDACs activity when we treat the cells with TSA and culture before lysate preparation. Please suggest.

HDAC/Histone deacetylase seems to be interesting in differentiation. Further, Histone H3.1 is also interesting, since Histone H3.1 is the top major protein in fetal developing Hc cells (please see file; HepG2 Fucoidan).
Therefore, I have searched for these proteins in my protein database.
Fetal hepatocyte Hc cell has Histone deacetylase 4/HDAC4 at 15.0 μg/mg of cell protein, and Histone H 3.1 (1-136) at 49.4 μg/mg of cell protein. Therefore, these important proteins are working at 6.44% among cell proteins.
Hepatoma HepG2 (cultured without fucoidan) has Histone-lysine N-methyltransferase SUV39H1/Histone H3-K9 methyltransferase 1 at 9.7, and Histone H 3.1 (2-136) at 6.3 μg/mg of cell protein, respectively.
Healed hepatocyte HepG2 (cultured with fucoidan for 3 days) has Histone deacetylase 7a/HD7a/Histone deacetylase 7/HDAC7 at 0.09, Histone H3-K9 methyltransferase 4/Histone-lysine N-methyltransferase, H3 lysine-9 specific SUVH4 at 0.67, and Histone H 3.1 (2-136) at 8.8 μg/mg of cell protein, respectively.
Serum of GSD-1b patient (with biotin deficiency and eplepsy) has Histone-lysine N-methyltransferase MLL3/Lysine N-methyltransferase 2C at 9.3 μg/mg of serum protein.
Serum of 4mo boy (with biotin deficiency) has Histone acetyltransferase MYST3/Histone acetyltransferase KAT6A; at 11.3, and Histone-lysine N-methyltransferase 2B/Trithorax homolog 2 at 4.5 μg/mg of serum protein, respectively.
Healthy serum of 33y male has Histone acetyltransferase KAT7/MYST2 at 7.7 μg/mg of serum protein.
LC tissue (with leprosy) has Histone H3.1 (1-136) at 11.9 μg/mg of tissue protein.
HCC tissue (with PBC) has Histone H3.1 (1-136) at 2.0 μg/mg of tissue protein.
HCC tissue (named as No.6) has Histone H3.1 (1-136) at 1.9 μg/mg of tissue protein.
Cultured hepatoma and healed hepatocyte HepG2 cells have uniquely 2-136 of Histone H 3.1. Establishment for cultured cell may changed to cut the N-terminal Met.
Histone deacetylases may be required for normal immortal cells to grow and develop.
By the way, I have mentioned PDMD method is suitable for your study to perform.
I have long studied about multiple hydrolase biotinidase and lipoamidase (please see files; Enk serBIN and Multiple Hydrolase LIP). I am now considering that biotinidase is biotinylation enzyme to carboxylases' -MKM- of K(Lys) residue as reversed reaction of biotinidase (please see file; Biocytine synthetase).
Histone deacetylases (HDACs) may have a multiple hydrolase characteristics similar to biotinidase and/or lipoamidase.
