I am trying to follow the proliferation of LnCaP cells during various treatments. This human prostate cancer cell line grows as adherent, single cells and loosely attached clusters. Therefore it is an especially difficult cell line to assess growth rate.
Sulforhodamine B was unsuccessfull. Does anyone has any method other than trypan blue? Should we coat the plates?

You can add WST Reagent (roche) directly into the media and measure a colour change via spectrophotometry.

It depends on what you are trying to measure. If you just want rates of proliferation, a standard population doubling assay is easiest. For measuring where the cells are in the cell cycle at any given time, then flow cytometry is easiest.
