Hi everybody. Does anybody know the maximum length of DNA (between left border and right border of plasmid) which can be transformed by Agrobacterium tumefaciens?
Virtually unlimited, but that of course would depend on the particular conditions (amount of expressed Vir proteins, infection time, etc), but you can even introduce BACs through Agrobacterium-mediated transformation:

Dear Can you please provide me in detail step by step protocol for Agrobacterium mediated transformation in Ganoderma lucidium. I will be highly obliged. Kind Regards
dear Ali there are many protocol for gene transformation to fungi but we only used C. Peter Romaine and Carl Schlagnhaufer method (Mushroom (Agaricus_bisporus)) for gene transformation into the common white mushroom.
Consider it unlimited although not true for A. tumefaciens and A. rhizogenes but transformation happens through 2 steps. First Agrobacterium mediated transformation into eukaryotic cell then its integration into genome. So the 2nd step would really be rate-limiting in context to integration into the host genome.
