I'm going to use half-strength MS media for root initiation for the indica rice cultivar. What will be it's composition when I'm using MS salt, sucrose, and agar without growth regulators?
You should double-check the literature about this 1/2x strength MS medium for your specific rice variety. Does the suggestion of 1/2x strength medium mean that the MS salts is 1/2x and so is the sucrose (1/2x)? In general, it also means 1/2x sucrose. But, to make sure, you should check. The agar should stay the same. Agar is only used to support the plants.
Yes; as suggested by Prof @Yuan, better double-check on the 1/2x strength of your rice line. For me, I used Upland rice and considered the1/2x strength as recommended above by Dr @Sam. Rather the sucrose I used was 10g/L.
You can reduce the concentration of mineral salts (macro, micro, iron) and organic (vitamins) of MS medium to half and decrease the amount of sucrose and agar to 20 and 6 g/L, respectively.
Different definitions as per experiments (a) 1/2 MS means 1/2X macro & microsalts while keeping the organic components, iron and agar unchanged. (b) 1/2X salts, organic component, iron while keeping the sucrose and agar unchanged (c) 1/2X of each components along with reduced sucrose content also.... Which definition suits better to the term 1/2MS?
Half strength of MS medium means the use of half the quantity of macro and micronutrients including sucrose. The gelling agent will remain in the same quantity.
