I isolated a virulent Salmonella phage from water, and I want to preserve this phage until I complete the screening for more Salmonella phage. I want a detailed method to preserve this phage for along time about four months.
Depends on the kind of phage the storage length is so different. But you could store your plaque on the LB broth with 20% glycerol at -20 for few weeks. And also if you put it on the SM buffer you could store it at 4c for few days. In some literatures, it is mentioned that freeze-drying could be effective to store phage for long time.
We store EMBL3 phage plaques in SM bufer (SM Buffer (per Liter)
5.8 g of NaCl, 2.0 g of MgSO4 · 7H2O, 50.0 ml of 1 M Tris-HCl (pH 7.5), 5.0 ml of 2% (w/v) gelatin. Add deionized H2O to a final volume of 1 liter. Autoclave) with gelatin over 5 years in -70oC. http://www.chem-agilent.com/pdf/strata/241211.pdf .
