I'm using commercially available kit and plastic plates, but I heard that maybe wells coated with polylysine would provide better results? Does anyone have any experience with osteogenic differentiation on plates coated with polylysine etc?
Hi Patrycija, these are our 2 established protocols for osteogenic differentiation, you need to change them slightly to adapt to your needs but I have been using them for a long time and they work really well in our lab. hope they help you
Antonio M Spadaccino For the protocols detailed above, would they work for the osteogenic differentiation of bone marrow derived MSCs as well as adipose derived MSCs? Many thanks!
I think so, normally the protocol works very well with bone marrow derived sc so i don't see why should not work from adipose derived sc. the only thing i would do is try anyway, the reagents are quite easily found in any labs. The only thing i remember was the difficulty of isolating sc from adipose tissue and not the differentiation. I abandoned it at the time because was very low yielding and did not allow to do many studies from the lack of sizeable amount of cells found
