I am finding the optimum temperature and pH of thermostable enzyme and its mutants. I find the optimum temperature of wild type enzyme and its mutants as 80C and 70C at 10 minutes incubation. but for thermal stability when I incubated the wild type enzyme at three different temperatures as 70,80 and 90 degree centigrade for 4 hours incubation , I find that the enzyme completely lost its activity after 10 minutes incubation for 90C and 80c of 4 hour incubation while for 70c it gives uniform decline in activity with time .

Please let me know why the enzyme is losing its activity at its optimum temperatures ? The enzyme was showing optimum activity at 80C, when I was finding its optimum temperature but when I used this 80C temperature for its thermal stability at four hours incubation without substrate then I find that up to 10 minutes it showing activity but after 10 minutes it completely lost its activity .

can any one tell me the reason or any suggestions for using other methods for accurate optimum temperature and thermal stability ?
