I am interested in looking at the ROS in the cytoplasm and mitochondria using some ro-GFP probes for which I have to transfect primary hepatocytes with these plasmids. I have already tried Liposomes for this but it is not working as expected. Literature (very few papers available) says it is the best method, but I just wanted to ask whether anybody has some experience with liposomes or any other transfection method for transfecting primary hepatocytes which works best?
Transfecting primary hepatocytes is indeed very challenging. The most efficient will be to use a viral system. However, there is a magnetic nanoparticles based transfection technology called Magnetofection, that has been very successful in numerous primary cells and hepatoma, see http://www.ozbiosciences.com/magnetofection.html

I would agree with Olivier. The most effective way for many primary cell types are usually the viral based methods usually adenovirus based (i.e.there is a kit called cytofect which has been optimised for hepatocytes). good luck with your experiments.
