I need to extract RNA from islets of Bl6 mice. So I was just wondering which is the best kit for extracting RNA from a low number of cells (around 100 islets only)? If anybody has experience with islets please do share!
I prefer the Trizol (or Trireagent), used according to the recommended protocol, resuspending the pellet in a smaller volume of nuclease free water (or DEPC treated water); adding 1 ul of LPA (Linear polyacrilamide) in the precipitation step increases the amount of RNA, particularly for the small molecules. The Qiagen has indeed an RNeasy micro kit, dedicated to small volumes but without DNase treatment the amount of DNA contamination is higher. Additionally, being a column - based kit, the loss of RNA is higher than with a precipitation method. As mentioned previously, the homogenization step is very important, the instruments using a rotor-stator being the most efficient.
Hi, Qiagen is a good kit and you can do some modifications, also there is a mini kit if you don't need a lot of reactions colone, also Roche is a good one and the area of modifications is permitted more, regards!
