In order to extract DNA from Streptococcus from pure culture which method is recommended. I found many manual extraction methods, some recommend use of mutanolysin/ lysostaphin for the better yield. If we are using kit based method which one is better. Qiagen provides a range of extraction kit(QIAamp mini kit/ DNeasy kit/ mericon bacteria plus kit) which one is best suited for my purpose. Can i go for HIMEDIA extraction kit? Can anyone help me?
Qiagen DNeasy tissue kit ........ so good,
minor modification: to increase the DNA concentration of the final yield, divide the elution buffer into two volumes (50 microliter each), add the first volume on the filter of the column and incubate at room temperature for at least 30 minutes, spin as described, and add the other volume of elution buffer again with 15 minutes incubation then spin, so total volume is 100 microliter stock DNA with high conc.
Collect bacteria directly from the plate, better results that broth suspension

If you do not need to quantification, it is not need the manual extraction methods. I think any bacteria Kit is OK for this bacteria DNA extraction

QIAGEN or Cal Biochem, best kits for high purity and yields.
Qiagen DNeasy extraction kit is very nice and you can add lysosome also it can hydrolyzes glycosidic bonds of peptidoglycan in Gram-positive bacteria. so you can get better yield.

If you want the cheapest method is enough with boiling a suspension of the bacteria in 300ul with destilated water or saline solution, boil during ten minutes. After that centrifugate the sample at 3000rpm per 5min and use the supernatant to do the PCR
