I am working on the molecular epidemiology of beta haemolytic Streptococcus. Although I have the CDC guidelines for emm typing, I am confused in procedure especially after PCR step. Can anyone help me? Can anything new can be done in Beta haemolytic Streptococci?

after PCR we purify the emm-amplicon with the QIAquick PCR Purification Kit
(Qiagen, Hilden, Germany) before it is subjected to Sanger-sequencing. The
emm-type is then assigned by BLAST comparison with the emm-genes
of the CDC database (http://www.cdc.gov/ncidod/biotech/strep/
M-ProteinGenetyping.htm). If the homology to the database sequences is low you may have discovered a new emm sub-type or type and should contact the curators of the CDC-databese for help.
