I am starting a project that requires to induce a chronic systemic inflammation to mice. I try to figure out which dose of LPS I can give to mice and maybe how many times I should give them to ensure they have chronic systemic inflammation.
I saw data in a few articles on Pubmed but I don't manage to find a compromise between the dose and the frequency... the point is we aim at inducing denutrition through systemic inflammation.
Any ideas ? thanks

LPS responsiveness varies between mice strains and animal house hygiene. To induce systemic inflammation a single i.p. dose of 100 microgram LPS per mice should be enough. You need to do a pilot experiment to optimize the LPS concentration in the experimental paradigm.
Hi, thanks for your response ! Indeed I am doing a pilot experiment in which different doses of LPS are used to mice and then observe survival rates. I am using C57BL6 adult male mice.
The best situation would be having the mice in chronic inflammatory state for a long time enough to see them in undernutrition.

Hi and many greetings, I am working currently on developing systemic chronic inflammation model in SD rats. Indeed, I would like to advice you to measure also the food intake, body weight and body temperature, as long as LPS would affect on the above-mentioned physical features. In addition, you can try to observe the developing of chronic inflammation through measuring neutrophils and monocyte or macrophage where the latter one increase steadily through development of chronic inflammation.
With respect to your area of interest (denutrition), using LPS would cause initially anhedonic effect and reduce the food intake. However, LPS tolerance in mice could return the food intake to its normal levels. Therefore, you need to revise the previous published articles (LPS and food intake) to choose the most suitable dose and administration method (pellet or i.p injection). Good luck to you :)
