Phytosterols can be extracted using petroleum ether or hexane as they are non polar in nature. Dichloromethane and chloroform are also good ones but initial extraction should be with hexane or petroleum ether preferably. You can use a soxhlet extractor for this purpose.

For HPLC method....u can simply use methanol water combination 40:60... or 60:40 as per ur Rt requirement

For HPLC of phytosterols, methanol:water (80:20) is best and proved successful in most of the cases.
