Ideally blocking has to be done at 4 degree. Since BSA or milk is rich source of nutrition for bacteria and other things. So better be on safer side. However if blocking is just for an hour or so, then it is ok.
As someone already said, if you are blocking overnight, it's better to do it at 4ºC. Otherwise, 2h at room temperature it's normally enough. Also, we always block the membrane with gently agitation (using the roller, if you keep the membrane in a tube, or the rocker, if you keep it in a weighing boat) to ensure complete blocking.
Yes I think so. With my experience, room temperature is the ideal. But maximum should be around 6-8 hrs in room temperature. But if we want to keep longer, it is better to keep at 4 degrees. When temperature is decreased, incubation time should be increased.
