Hello everyone,
I need details about mesenchymal stem cell (bone marrow stem cell) media which can be used for clinical trial studies and which is clinically approved.
Our application can also involve the injection of this media into patient. I need media which is safe for human applications and properly approved clinically.
Please add your suggestions.
There are a number of companies which provide Xenofree MSC media, you shuold look into what is commercially available.
Thanks for the answer Oscar M pello. but the mentioned media in cited refrence is a replacement for supplements and FBS. we are already using human Platelet plasma to replace FBS. What I am asking is a culture media which can also be injected in human, not a replacement of supplements. Thanks for your time and logical suggestion. Any suggestions regarding culture media dear Oscar?
Thanks DONG, But I think DMEM-F12 is not a good choice if we talk about human applications,,,it is a general proliferation media..
Dear Oscar, we are not planing to infuse MSCs....Secondly, if we can use fresh MSCs then why to go for cryopreservation, because efficacy and homing in property of MSCs is reduced in that case... Fresh MSCs can be infusd into normal saline and already such kind of clinical trials are going on here but our protein of interest is secreted in culture media..so we have to infuse media only
I recomend alfa mem. Usually the manufactorer informs if it is suitable for clinical trials, according to the prodution criteria and absence of phenol red and other substances.
Thanks Carolina.. I use alpha MEM from Sigma in powder form.. Which media are you talking about actually? Kindly tell company and cat. no. so that I can have a look on the same!
At the moment I am in pre-clinical research, so I am not using any. But in the MSD the manufacturer informs if it is suitable for clinical research or not. I didn't find any in the Sigma catalog, but I found several culture media classified as suitable for clinical research in the Gibco and Cutilab catalogs. You just need to find the most appropriate for your cells and classified as clinical compatible. Sorry, I wish I could be more helpful.
Life Technologies makes a xeno free Stempro MSC-SFM medium. Since this is serum and xeno free, this should be suitable for clinical trials.
you can use DMEM from Lonza which is specified for clinical use not research.
Also there are some FBS from PAA company or Worthington which are extracted from non cow madness endemic areas as Newzeland or Australia for your seru suplement or autologous serum.
As trypsin substitute you can use TrypLE as a GMP grade enzyme
Ficollpaque from GE as a GMP grade ficoll
also you can use clinical PBs for washing procedures named as Clinimax for clinical application
try to wash cells before injection because remaining phenol red can induce flushing and some minor allergic reactions
