I have used individual antibodies from many sources (Sigma, R&D, Jackson, KPL, AbDSerotec, eBioscience, and others) with good results. First, determine if the antibody is truly specific for the product you wish to measure. Second, when possible get a labeled detection antibody to avoid the extra noise of double sandwich assays. Then, test every thing with internal positive, negative and dilution standard runs to assure you know the limits of the test. ELISA is not only about buying antibody or kits blind. KNOW WHAT THE TEST MEASURES in quantity and quality.

I've had very good experiences with R&D Systems so far.

Although I have not used more than a few vendors for antibodies, I have been most pleased recently with Bethyl Laboratories.

Whichever antibody best works for your assay. You should get multiple vendor antibodies, if available, and look at specificity and sensitivity in your assay.If it is going to be used in a Regulated assay be sure the vendor can provide you product through the project and that they have documentation on antibody production. What QC are they using when releasing the antibody.

I have good experience with antibody pairs from Mabtech and R&D Systems.

We use R&D Systems unless they don't have the antibody pair in the compound and species of interest and have been happy with the results. We titer the antibody pair when we they arrive in the lab for optimal sensitivity and dynamic range. We re-titer every 2-3 months and when/if the titers fall below 1:250 we buy a new set.

In my view invitrogen antibodies are the best and there is no need to think about others

If you are doing human serum/plasma R&D have high sensitivity ELISA.

I think the antibodies which worked well before in your hand should use. We are using antibodies from Southern Biotech from last 10yrs and never had any problems.

We use R&D and Jackson. I agree that you need to try a few and see what is best for your assay. Also your combination of monoclonal and polyclonal and the host of antibody ie rabbit, goat etc will make a big difference.

I would go with Abcam, R&D, Peprotech, BD.

It sometimes depends on specific situation to prefer one company to another.

Price considerations can kick in as well.

It depends on the species you are interested in. R&D offers quite reliable antibodies for ELISA . They also offer ELISA kits for both large animal and mouse research.

Hey, antibodies are not mobile phones. Most brands differ just in ridiculously high prices. Most antibodies which are commercially available are re-sold and still originate from research groups who sell their surplus materials. Actually, for the price you spend for a few 100 micrograms, you already may have developed your own in quantities if you can supply an antigen and dare to wait for 2 or 3 month :)

Of course, any brand will give you antibodies of your interest. But, in my opinion preparing your own antibodies against any antigen is the best method. Raise antibodies and you can compare them with any brand and most important thing is comparing with reference antibodies. So, better to raise your own antibodies and subject them intra and inter laboratory validation and comparing with reference antibodies is always good.

I have used a few different brands to develop ELISA assays including abcam, genway biotech and other antibodies which have worked very well. They are of course expensive so if you can find something in the literature where a pair of antibodies has been used this will save you some trouble.

I have had most success with BD Pharmingen, eBioScience and R&D antibodies. If you stick to matched antibody pairs from these brands, you should have no problems.

Antibodies are proteins that recognize unique parts of foreign targets /antigens and react specifically with and bind to the antigens. So you have to ask yourself basic questions:

1. What are the procedures (methodology) that you would want to employ?

2. What are the reagents or antibodies that you need and against which antigens

3. If you want to detect antigen X, then you need antibody against X or anti - X.

If you need to detect antibody Y, then you need anti-antibody Y

4. If you need to detect a single epitope, then you require monoclonal antibodies ,

but they are more expensive than the polyclonal antibodies.

So there is no brand that is superior to others. Antibodies are manufactured to the requirement of the end users for the purpose of detecting specific antigens. If your requiment is to have high sensitivity and specificity, but with low background noise, then you have to purifiy them according to your needs. Purified antibodies will be a little bit more expensive.

We use R&D and work usually well. The standards we usually make 10x stocks, aliquoted in 50µl so we have plenty. The cost can be cut down if you use half of everything per plate.

They are able to recognize much more than the antigen described in the catalog. Often are repetitive antigens here like ssDAN, dsDNA, collagen, cytokeratins, .... Just check this.

As secondary (laballed) antibodies are some really excellent suppliers: www.seramun.de is one of the best. The polyclonal antibodies have a excellent specifitcty, the labeling is stable and in a high molar ratio so that you can use low concentrations.

I used a lot of eBioscience antibodies for ELISA, ELISPOT, and flow cytometry, with very good results. It depends heavily on your species of interest and what types of antibodies you require (and who can supply them).

Unfortunately nobody mentioned the PeliKine kits from Sanquin yet, since I am from Sanquin my selves I realize that it is not appropriate to post an answer however all the names of other brands have been mentioned already. Therefore, you had best find out your selves by google (scholar) on -PeliKine- or Lucien Aarden to find the origin of the antibodies.

First what antibodies do you require? I mean against which antigen? In my opinion, prepare your antibodies after purifying antigen in question and after checking the purity of antigen. Then, compare the efficacy of your antibodies raised in suitable lab animal or host species with reference antiserum / antibodies. If your antigen is pure and if it is not having any difference with reference antigen strains, then certainly the antibodies raised by you are of best quality. Instead of purchasing antibodies better you raise and check antibodies, definitely which are of good quality.
