I've been using 96-well plates for growing cells under exposure to ethanol, but I'm also aware the multi-well plates can have evaporation issues. I'm wondering if there is a difference in the ethanol evaporation when using 96-well vs. 48, 24 or 6-well plates, and if so, which is the best one to use.
Hi Jeremy. I think regardless of the well-plate that you use, evaporation is going to be always an issue. I've found this technical bulletin comparing the evaporation among different plates:

For your assays, try to use plates with lids with edge rib and rings, and if possible work in an incubator that no other people would be using (opening and closing it several times causes disturbances in the humidity of the incubator). As you'll find in this reference, the wells in the "periphery" have more evaporation as those ones in the center. If you work with 96-well plates, you can use only the central ones, and fill the ones in the extremes with sterile water (200µl/well).
