Hi, I'm an undergraduate student trying to isolate Pseudomonas putida from the soil for a project. I'm wondering what is the best media that can be used. Is it the Kings B agar or the Cetrimide agar? Thank you in advance for your help.
Tengku Zalia Sofea Tengku Zainal Azman According to the original sources, Cetrimide agar agar is King'sB + сetrimide. Although, some other basic recipes for nutrient agar + cetrimide were tested as well (Brown VI, Lowbury EJ. Use of an improved cetrimide agar medium and other culture methods for Pseudomonas aeruginosa. J Clin Pathol. 1965 Nov;18(6):752-6. doi: 10.1136/jcp.18.6.752). So, King's B is just non-selective variant of cetrimide agar.

It will still be difficult to differentiate putida from other Gram Pseudomonas spp. Cetrimide was developed for clinical isolates - some Pseudomonas spp. may not grow..
