May I know if anyone have experience in culturing UC-MSC into organoids using Matrigel.
I have dissected Mouse lung, dissociated them into single cells and added Matrigel, hoping that it will reorganise into lungs with the help of UC-MSC. Unfortunately, cells could not form organoids, instead remain attached to the plastic. My control showed formation of a decent size organoids.
Please if anyone know how can I troubleshoot? I hypothesise that by co-culturing UC-MSC with dissociated native lung cells can stimulate organoid formation, however this is not the case.

You might consider increasing the number of MSCs. The balance (cells and matrigel) is important to creating organoids.
Using special culture plates to create spheres would be the other approach. The following paper (Figure5) may be helpful.
