I am performing MTT assay on PBMCs. For adjusting cell number, Briefly I seed different number of cells/well, incubate 24 and 48h, add MTT and incubate, finally add DMSO as solvent and read the plate at 570nm and 630nm as reference.

My question is that what is the appropriate wavelength in MTT assay? I see different wavelength in different articles. Considering that UV-spectrum of MTT formazan is approximately from 450nm to 650nm, is it right to consider 630 as reference while it still is in UV-spectrum of MTT formazan (450-650). Isnˈt it more accurate to consider reference OD higher than 650nm?

I have low ODs when I read the plate at 570nm, but good absorbaance at 490 (Considering that maximum abs of MTT formazan is at 490-500nm ). Can I use 490nm or it is not accurate. If it is not, please tell me the reason.
