What is teh best way to differentiate 5-NT activity form alkaline phosphatase activity?

For differentiation between 5-NT and alkaline phosphatase activity, is it best to add nickel or concanavalin A ti inhibit 5-NT, or is it better to add ALP substrate beta glycerophosphate?
