I want to know what is the incubation period after injecting 263k infected brain homogenate to Female syrian golden Hamsters? When do signs appears? what are first clinical signs? and when do they die usually?
I completed a response for you based on intracerebral route of inoculation before taking in your add-on message that you wanted answers for the intraperitoneal route. Sorry that I cannot advise on ip, but I hope you will find parts of my already prepared answer still relevant. I have deleted the paper I attached and also cut out parts of my answer that referred to ic.
I can tell you what we see in mice so that you can infer from that. The difference between IC and IP routes in wild type mice challenged with RML in our Lab is about 50-70 days (it is not double. We have 70 days incubation period for 263K ic inoculated, but we do not have ip data. You may be able to take an educated guess from this if you cannot get exact data from any other source.
We do not allow inoculated animals (hamster or mice) to die from prion disease, because we are bound by our project licence to cull at a maximum of moderate severity, which is before the animals suffer substantial adverse effect. Only 5% of animals are allowed to die rather than being killed at the experimental endpoint just to account for instances beyond out control (e.g. animal dying overnight). In one of our experiments that I have just referred to, hamsters challenged with 263K ic are killed within 5 days of the first signs that appear around 70 days post inoculation that include: Piloerection, Ungroomed Appearance, Erect Ears, Marked Aggression. Ataxia would usually be diagnostic and would trigger immediate termination. The diagnostic sign will be similar and the incubation period will of course be much longer when you use the i.p route. I hope you do get a realistic incubation period for ip inoculated 263K into Syrian golden hamsters from another source.
thanks alot sir for such a nice reply...and u know there are confusing papers...one says ip rout gives quicker signs at 60 plus days and other says 90 so im not sure but at least im observing my animal closely so i hope i can answer my questions myself...thanks again
