Any simple lysis buffer that realses total cell content would be useful. We often use RIPA buffer (radioimmunoprecipitation assay buffer) and it works well..

the tissue need to be homognised in KCl 1.15 % 

You can use sodium chloride Nacl normal saline. You can add 1 gm tissue to 9 ml saline to be 10% concentration

I used Tris-Base 20 mM buffer, pH=7,6:

EDTA 1mM

Tris-Base 20 mM

KCl 150 mM

Sucrose 500 mM

-mercaptoetanol 1 mM

Homogenize with teflon pestle and centrifuge cold at 14,000 X g for 10 min. Save the supernatants can be stored at -20 ° C or -80 ° C.
