When an enzyme is incubated in a temperature higher than its optimal, what physical and chemical changes happen within that protein?

Change in the three dimensional conformation leading to loss of potential binding site on the enzyme. This might affect the kinetics of the enzyme reaction (not to mention Km Vmax).
