I am working on cotton, and I did successfully silence a particular gene using one of the available commercial vectors. The success of the VIGS was over 99% as per the albino nature on the seedling leaf which also spread to the stem region. I wish to evaluate the functional role of the silenced gene on cotton fibre, what are the requirements in order to maintain the gene at silenced state, having in mind that as the plant grows, various changes occurs or do I have to keep on introducing the vector containing the silenced gene segment as the plant grows. Kindly any solutions towards this, being majority of the VIGS experiment are mainly done at the seedling stage as opposed to advanced stages of plant development.
