I'm working in an industrial field trying to improve some products and amongst them is Giemsa stain. The problem is, our solution cannot stain chromosomes of peripheral blood cells but control slides of other companies' solutions do. What could be the reason of the lack of staining in my solution?
If I understand the question, you stain your preparations in duplicate using "your" Giemsa and another manufacturer. Is that it?
Do you prepare a solution from commercial Giemsa powder or produce the Giemsa powder as a mixture of methylene blue-eosin-azur?

I am using a Giemsa solution provided by Merck and a solution mixed at our own facility from methylene blue, eosin and azure b and c.
However I managed to get the stained chromosomes by adding a bit of acetone in the mixture :)
I summarize what I realized:
- you use a modified Giemsa Merck with the addition of acetone
- at the same time, you stain both your slides and control slides
- the result is: no coloring of your slides; colouring of the control slides. Is that it?
If you confirm these steps we should think that the problem is not in Giemsa but in the preparation of the samples. What are the samples? If they’re metaphases, how did you get them?
