I want to perform antioxidant assay using DPPH solution. Is it fine if the wax solution is a bit turbid for DPPH assay. Since, turbid solution does not work for antimicrobial assay for me.
Try DMSO, and make a high concentration. Finally, dilute it with water and perform the assay. Always put a same dilution DMSO as control to eliminate the chances of false positive result. Hope that works.
You could try to dissolve your extract in DMSO and use one DMSO control in your anti-microbial study. I also would suggest to check extract solubility in different percentage/ratio of DMSO in saline.
Instead of DMSO, you also can use Polyethylene glycol (PEG) or Tween-20.
We tried a variety of solvents ranging from chloroform, acetone, DMSO, methanol, hexane etc.Actually the solubility of wax in a particular solvent depends on the composition of wax.
For DPPH assay, we continued with methanol for our waxes with a bit of heating.
Please note the waxes are a bit difficult to work with and you may never find 100 % solubility in any solvent. You may have to check more than one solvent, different dilutions.
