I have read here and there that upregulations of the proteins BiP and CHOP are considered as markers of ER-stress. However in my case, I see severe down-regulations of these proteins. Could it also be linked to impaired ER function?
If you do a quick image search for UPR you will see that CHOP and Bio (aka Grp78) are some of the final steps in this stress pathway. And, as such, they (particularly CHOP) really shouldn't be high to start with. CHOP is basically an apoptosis signal, and in cell lines I work with it is barely detectable without causing ER stress.
May I ask what you are doing to get this response? Treating with a compound? My guess would be that you're somehow stressing your negative control. Now if you really do have high CHOP levels, it's possible that your treatment is allowing GAD34 to get activated by CHOP which will then negatively regulate the whole pathway.
I'm sure this isn't terribly helpful, but maybe with a little more info about the treatment perhaps I (or others) can provide more assistance.
