When you purify human monocytes from PBMCs (magnetic selection) we have noticed some differences in the brand of tissue culture plates used. Has anyone compared different brands like BD to Corning to Sarstedt to Costar for monocyte survival and adherence? We only need to culture for 24 hours and are playing around with densities in 12, 24 and 48 well plates to maximize survival with various conditions. Do you let them rest and adhere for an hour after isolating them before adding stimuli? We currently use 10% FBS in RPMI. Not all monocytes seem to attach but I'm guessing there must be a 'magic' density to keep them happy. Thanks
