I am designing an intracellular cytokine staining panel to assess TLR-induced type 1 IFN responses in human blood monocytes. I'm not sure if I should try staining for IFN directly, or some IFN stimulated genes like CXCL10 or an MX protein. I'm sure this will also depend on my stimulation time with TLR agonists, which I'd like to keep at around 4-8 hours if possible. Any insights into how best to set this panel up would be appreciated.
