I have been looking at Th17 cells. I usually stimulate my PBMCs with PMA,ionomycin and monensin, perform surface as well as intracellular staining for Th17. Gate down CD3+CD8- cells (mainly Th, few NKT also) to RORgt+IL-17+ cells. Sometimes in patients what I usually see that the %of RORgt+IL-17- population is higher in comparison to healthy control. I have been buying RORgt from ebiosciences, other then Th17 cells the receptor RORgt also expresses on CD4+CD8+DP and other then RORgt the Ab shows binding with RORg also. The Th17 cells what I usually get shows IL-17 bright and RORgt intermediate expression. IL-17 no doubt is the best for Th17 characterization.But still I'm eager to know why these cells show intermediate expression of RORgt and what are the cells those are IL-17- and shows brighter expression of these transcription factors (RORgt/RORg)? I would be thankful if somebody give me more information on RORgt.
